Protein-L-isoaspartate O-methyltransferase domain containing 1 (PCMTD1) is a mammalian protein characterized by the presence of an N-terminal domain similar to l-isoaspartate and S-adenosylmethionine binding motifs, implying structural similarity to PCMT1. Unlike PCMT1, PCMTD1 does not catalyze methyltransferase activity under tested conditions. Instead, it is proposed to function as a substrate adaptor for Cullin-RING E3 ubiquitin ligase complexes by binding to damaged proteins containing l-isoaspartate and facilitating their recognition and possible proteolytic degradation. The protein contains a SOCS box motif responsible for E3 ligase recruitment. PCMTD1 thus forms part of an alternative protein quality control system in mammalian cells, supporting maintenance of protein integrity by targeting damaged proteins for degradation, rather than enzymatic repair.
