Overview

Protein mab-21-like 3 (MAB21L3) is a member of the highly conserved MAB21-like family of nucleotidyltransferase-fold proteins that share structural homology with the cyclic GMP-AMP synthase (cGAS), a sensor of cytosolic DNA in innate immunity[1][2][4]. MAB21L3 is less characterized than MAB21L1 and MAB21L2, but is implicated in cell fate determination during development, especially in the context of multiciliate and ionocyte cell differentiation downstream of Notch signaling in vertebrates[2][4]. The exact molecular function, biochemical activity, and ligand interactions of MAB21L3 remain unknown—unlike cGAS, its nucleotidyltransferase activity has not been demonstrated and may require conformational change for activation, or may be absent[1][2][4]. Mutations in closely related proteins (MAB21L1, MAB21L2) are associated with severe developmental disorders, highlighting the likely crucial, though incompletely defined, role of these proteins in vertebrate development[1][2][4]. No known drugs, biomarkers, or disease associations specific to MAB21L3 are described in the current literature.
