Overview

Protein mono-ADP-ribosyltransferase PARP8 (PARP8) is an enzyme belonging to the poly(ADP-ribose) polymerase (PARP) family, specifically classified as a mono-ADP-ribosyltransferase (MART). Unlike the best-known PARP family members (such as PARP1 and PARP2) that play established roles in DNA repair, PARP8's physiological function is less well characterized. PARP8 localizes predominantly to the nuclear envelope in mammalian cells. Knockdown studies suggest it is critical for the organization and integrity of the nuclear envelope, influencing nuclear morphology and potentially nuclear envelope biogenesis[2][4]. Additionally, there is evidence for its involvement in the assembly or maintenance of membranous organelles[3][4]. PARP8 uses NAD+ as its substrate, modifying target proteins by the addition of ADP-ribose moieties (a process known as mono-ADP-ribosylation)[3][5]. As of now, PARP8 is not implicated in major human diseases nor targeted by therapeutic drugs, and its direct roles in signal transduction, DNA repair, apoptosis, or other cell signaling pathways remain underexplored.
