Protein N-lysine methyltransferase METTL21A (METTL21A)

Target
METTL21A
Molecular classification
Enzyme, Protein-lysine methyltransferase, Protein methyltransferase
01

Overview

METTL21A (Protein N-lysine methyltransferase METTL21A) is an enzyme that selectively trimethylates lysine residues in members of the heat shock protein 70 (HSP70) family, including HSPA1, HSPA2, HSPA5, and HSPA8. This post-translational modification may regulate chaperone activity, protein folding, and responses to cellular stress. METTL21A has been associated with diseases such as pontocerebellar hypoplasia type 2A and malignant ovarian cyst. Its functional role focuses on modifying HSP70 proteins rather than directly mediating signal transduction or cell-surface signaling[1][3][4][5][7][8].

Mechanism of action

Methylation of HSP70 family lysine residues (no clinical drugs with established mechanisms targeting this molecule as of now)

03

Biological functions

Protein methylationPost-translational modificationRegulation of heat shock protein 70 (HSP70) activity
04

Disease associations

CancerNeurodevelopmental disorders (association with pontocerebellar hypoplasia, type 2A)Other (role in protein quality control and cell stress responses)
