Overview

Protein p13 MTCP-1 (MTCP1) is a member of the TCL1 family, encoded by the MTCP1 gene located on chromosome Xq28[5]. It is best known for its involvement in mature T-cell neoplasms, particularly T-cell prolymphocytic leukemia, where chromosomal translocations drive its overexpression[4][3]. MTCP1 adopts a unique eight-stranded antiparallel β-barrel structure and acts primarily as an activator/adaptor for AKT kinases, thereby promoting cell survival and proliferation[1][6]. The protein exists as a homodimer and interacts with AKT1/2/3 (through PH domain binding) and is also associated with mitochondrial membrane potential regulation[6]. MTCP1 has no known canonical ligand- or hormone-binding function and is not classically categorized as a receptor, enzyme, or channel. It is considered an oncogenic adaptor/coactivator involved in leukemogenesis and is a potential biomarker in certain mature T-cell malignancies. There are no drugs currently approved that directly target MTCP1, but its role in AKT pathway activation may inform targeted strategies in relevant leukemia subtypes[4][7].
