Protein phosphatase 1 regulatory inhibitor subunit 11 (PPP1R11) is a dual-function protein: it acts as an inhibitor of protein phosphatase 1 (PP1), a major serine/threonine phosphatase involved in diverse cellular processes, and it also functions as a RING finger E3 ubiquitin-protein ligase responsible for the ubiquitination and subsequent proteasomal degradation of Toll-like receptor 2 (TLR2)[1][2][4]. PPP1R11 thus plays an important role in regulating both cellular signaling by reversible protein phosphorylation and innate immune responses. Specifically, it inhibits the activity of PP1, and, by targeting TLR2 for degradation, it downregulates TLR2-mediated inflammatory cytokine production and bacterial clearance during gram-positive infections[2]. The gene is located within the major histocompatibility complex (MHC) class I region on chromosome 6 and has multiple established aliases, reflecting its study in both immune and reproductive contexts. No direct small-molecule drugs or clinical therapeutics currently target PPP1R11, and there are no validated clinical biomarkers or safety concerns directly linked to this protein.
