Protein phosphatase 1 regulatory inhibitor subunit 14B (PPP1R14B, also known as PHI-1) is a regulatory protein that functions as a potent inhibitor of protein phosphatase 1 (PP1, specifically the catalytic subunit PPP1CA), with over 50-fold higher inhibitory activity when phosphorylated[2][3]. It plays essential roles in regulating the cell cycle, apoptosis, and actomyosin contractility. PPP1R14B is strongly expressed in various tumor types and is associated with poor prognosis in cancer. Its expression correlates with immune cell infiltration (notably myeloid-derived suppressor cells) and pathways including cell cycle and RNA processing, suggesting fundamental roles in cancer progression and immune regulation. PPP1R14B serves as a prognostic biomarker in several malignancies and is being explored as an antitumor target, but no approved drugs are known to directly interact with or target this protein[1][2][3][5].
