Overview

Protein phosphatase 1 regulatory subunit 17 (PPP1R17), also known as G-substrate or GSBS, is a protein primarily expressed in cerebellar Purkinje cells where it functions as an inhibitor of protein phosphatase 1 (PP1) and protein phosphatase 2A (PP2A)[2][7]. It is a substrate for cGMP-dependent protein kinase, and its activity modulates intracellular signal transduction and regulates phosphatase activity—processes important in neural function and development[2][4][7]. Genetic variation in PPP1R17 has been associated with an increased risk of familial hypercholesterolemia and potentially with autism spectrum disorder[2]. There is currently no evidence that it serves as a direct therapeutic target (e.g., for small molecule drugs or biologics), nor is it a well-established biomarker or known to present notable safety concerns in therapeutic contexts.
