Overview

Protein phosphatase 1 regulatory subunit 18 (PPP1R18) is a protein that functions as a regulatory partner for the catalytic subunit of protein phosphatase 1 (PP1), a major serine/threonine phosphatase in eukaryotes. PPP1R18 directs the localization, substrate specificity, and activity of PP1 holoenzymes, contributing to a wide array of cellular processes such as cell cycle progression, mRNA splicing, and gene regulation. As with other PP1 regulatory subunits, PPP1R18 is critical for the finely-tuned, compartmentalized activity of PP1, making it a potential—but experimentally under-explored—therapeutic target. Dysregulation of PP1 complexes, through aberrations in regulatory subunits like PPP1R18, is associated with human diseases including cancer and neurological disorders. However, there are currently no clinically approved drugs that specifically modulate PPP1R18 activity or interaction with PP1.
