Overview

PPP1R35 is a regulatory subunit that interacts with protein phosphatase 1 (PP1), acting as a centrosomal protein required for proper centriole elongation and centriole-to-centrosome conversion. Its loss impairs centriole elongation and reduces centrosome number, highlighting a critical role in cell division and developmental processes. PPP1R35 functions via complex formation with RTTN (Rotatin), another centriolar protein, modulating the recruitment of structural and elongation machinery for centrioles. It harbors a conserved PP1-binding motif, but its centrosomal functions can be independent of direct PP1 interaction. PPP1R35 is enriched in vertebrate cells, and is suggested to play roles in ciliogenesis and centrosome homeostasis.
