Overview

Protein phosphatase 6 regulatory subunit 2 (PPP6R2) is a regulatory, scaffolding protein subunit for the catalytic subunit of protein phosphatase 6 (PP6), which is a member of the phosphoprotein phosphatase (PPP) family of serine/threonine phosphatases. PPP6R2 facilitates the assembly and function of the PP6 holoenzyme and is specifically involved in targeting the complex to certain substrates, such as those in the NF-kappaB signaling pathway and during cell division (mitosis). PPP6R2 acts to oppose the activation of the NF-kappaB pathway by supporting dephosphorylation of specific signaling intermediates, and loss of its function disturbs mitotic spindle formation and chromosome alignment. The protein is also known by several aliases, including SAPS2, KIAA0685, and SAP190. PPP6R2 mutations or dysregulation are implicated in Charcot-Marie-Tooth disease type 4B3 and may play broader roles in immune function and tumorigenesis.
