Overview

Protein phosphatase 6 regulatory subunit 3 (PPP6R3) is a regulatory and scaffolding subunit of the serine/threonine protein phosphatase 6 (PP6) complex, encoded by the PPP6R3 gene located on human chromosome 11q13.2. PPP6R3 (also known as SAPS3 and several other aliases) modulates the activity of the PP6 catalytic subunit (PPP6C) by determining its substrate specificity, recruiting specific substrates, and controlling intracellular localization of the complex. PP6 is involved in regulating essential cellular processes such as cell division, DNA damage response, inflammation, mRNA processing, and maintaining chromosome integrity. Disruption or aberrant regulation of PPP6R3, including rare fusion events (e.g., with USP6 in nodular fasciitis), can contribute to oncogenic transformation and tumorigenesis. PPP6R3-mediated complexes also regulate key effectors including Aurora A kinase (AURKA) and those involved in NF-κB signaling, among others. There are currently no drugs known to directly target PPP6R3, and it is not generally considered a druggable receptor, enzyme, or classical therapeutic target.
