Overview

Protein phosphatase inhibitor 2 (PPP1R2) is an intrinsically disordered regulatory protein that functions as a potent, specific inhibitor of protein phosphatase-1 (PP1), a major serine/threonine phosphatase involved in dephosphorylating a wide variety of cellular proteins[1][3][6]. PPP1R2 binds to PP1 through several conserved motifs (including RVxF, SILK, and the unique HYNE-IDoHA motif), blocking the PP1 active site and regulating its substrate specificity and activity within the cell[1][3][6][7]. Considered an ancient, evolutionary conserved PP1 regulator, PPP1R2 affects cell cycle progression, mitotic spindle function, and is necessary for proper centrosomal and chromosomal regulation in dividing cells[2][3][1]. While PPP1R2 is not itself an enzyme or classic receptor, its depletion or dysregulation leads to aberrant phosphorylation states and defects in cell proliferation, suggesting indirect relevance for cancer and other diseases involving disrupted cell cycle control[3]. No approved drugs target PPP1R2 directly; rather, it is a molecular regulator of the essential cellular phosphatase PP1[6][4][3].
