Overview

Protein PIGBOS1 is a small, 54-amino acid microprotein localized to the mitochondrial outer membrane, where it plays a pivotal role in regulating the endoplasmic reticulum (ER) unfolded protein response (UPR)[1][2][7]. PIGBOS1 interacts specifically with the ER-localized protein CLCC1 at mitochondria–ER contact sites, and its presence modulates cellular sensitivity to ER stress[1]. Loss of PIGBOS1 leads to heightened UPR activation and increased apoptosis, suggesting its importance for cell survival and homeostasis[1][2]. This protein is not classified as a therapeutic target or a member of traditional druggable protein families; instead, it represents a newly-appreciated class of microproteins essential for inter-organelle communication, but no clinical drugs, biomarker applications, or safety challenges are documented for this target at present[1][2][3][7].
