Overview

Protein polybromo-1 (PBRM1) is a subunit of the SWI/SNF (specifically PBAF) chromatin-remodeling complex, encoded by the PBRM1 gene in humans[1][4]. It contains bromodomains and BAH domains, enabling recognition of acetylated lysine residues—such as those on the tumor suppressor protein p53 and on histones—thereby regulating gene expression, chromatin structure, and genomic stability[3][6]. PBRM1 functions as a tumor suppressor, particularly in clear cell renal cell carcinoma (ccRCC)[1][2][5]. Mutations or deletions of PBRM1 are frequent in ccRCC and are associated with altered chromatin structure, deregulated gene expression, and distinct clinical outcomes compared to other tumor suppressor pathways[2]. PBRM1 contributes to the regulation of cell proliferation, cell cycle progression, and DNA damage response. Although not yet a direct pharmacological target, PBRM1 is an important biomarker, and its status can influence therapeutic response to immune checkpoint inhibitors and other cancer treatments[5][6].
