Protein prenyltransferases are a class of enzymes that catalyze the covalent attachment of hydrophobic isoprenoid groups (farnesyl or geranylgeranyl) to cysteine residues at or near the C-terminus of target proteins. This post-translational modification, known as protein prenylation, regulates protein localization, activity, and interactions, particularly for small GTPases. There are four main types: Farnesyltransferase (FTase), Geranylgeranyltransferase type I (GGTase-I), Rab geranylgeranyltransferase (GGTase-II/RabGGT), and Geranylgeranyltransferase type III (GGTase-III). These enzymes are therapeutic targets due to their role in diseases like cancer.
