Protein regulator of cytokinesis 1 (PRC1) is a non-motor microtubule-associated protein essential for central spindle formation and cytokinesis during cell division. It cross-links antiparallel microtubules at the spindle midzone, facilitating proper cell cleavage. PRC1 is tightly regulated by cell cycle-dependent phosphorylation and interacts with proteins such as Kinesin-4 and the centralspindlin complex. Dysregulation or overexpression of PRC1 is linked to genome instability, altered tumor immune environments, and poor prognosis in multiple cancer types, making it a candidate prognostic biomarker and a potential, though not yet clinically validated, therapeutic target.
Potential mechanisms for future drugs may include inhibition of microtubule bundling, disruption of PRC1 dimerization, modulation of PRC1 phosphorylation, and interference with its protein-protein interactions (e.g., Kinesin-4, centralspindlin complex)
