Overview

PORCN (Protein-serine O-palmitoleoyltransferase porcupine) is a multi-transmembrane enzyme located in the endoplasmic reticulum. It functions as a key regulator of the Wnt signaling pathway by mediating the post-translational attachment of palmitoleate, a 16-carbon monounsaturated fatty acid, to Wnt proteins. This lipid modification is essential for efficient binding of Wnt ligands to their frizzled receptors and subsequent signal transduction. Aberrant activation or dysregulation may contribute to cancer development. Mutations in PORCN cause focal dermal hypoplasia (Goltz syndrome).
