Overview

Protein shisa-6 (SHISA6) is a single-pass transmembrane auxiliary protein of the Shisa family, highly enriched in neuronal tissue—especially in the hippocampus and cerebellum—where it binds to and regulates AMPA-type glutamate receptors (AMPARs)[1][4][5]. SHISA6 modulates AMPAR trafficking, promotes their retention in the postsynaptic density by interacting with PDZ-containing scaffolding proteins such as PSD-95, and alters AMPAR channel kinetics by slowing desensitization and favoring sustained currents, thereby stabilizing synaptic transmission and supporting synaptic plasticity[1][2][3][4]. It is crucial for neuronal circuit refinement, memory formation, and may have neuroprotective properties. Mutational or knockout studies in mice show significant impacts on synaptic function and cognitive processes but SHISA6 is not yet a direct pharmacological target or disease biomarker[1][2][3][5]. It may also suppress canonical Wnt signaling in developmental contexts[2][6].
