Overview

Protein shisa-9 (SHISA9, also known as CKAMP44) is a transmembrane auxiliary protein that regulates short-term neuronal synaptic plasticity, primarily in the dentate gyrus of the hippocampus[2][4][6]. It associates with AMPA-type ionotropic glutamate receptors in synaptic spines, promoting AMPA receptor desensitization at excitatory synapses and modulating receptor trafficking and postsynaptic neurotransmitter receptor activity[1][6]. Protein shisa-9 interacts with several synaptic scaffold proteins and is dynamically regulated by Protein kinase C (PKC) signaling, contributing to synaptic plasticity mechanisms in the central nervous system[1]. Disease associations have been observed for epilepsy and autism spectrum disorder[2]. There is currently no evidence that SHISA9 is itself a direct therapeutic target for any approved or investigational drugs.
