A protein substrate refers to a protein molecule that serves as the target for enzymatic action, most commonly in biochemical reactions such as phosphorylation, proteolysis, or other post-translational modifications. In these contexts, the protein substrate is specifically recognized and bound by an enzyme (such as a kinase or protease), which then catalyzes a chemical transformation of the substrate. The interaction between an enzyme and its protein substrate is highly specific and arises from molecular interactions between amino acid residues. After binding at its active site, the enzyme catalyzes a reaction that alters the chemical structure of its protein substrate—either by adding/removing functional groups or cleaving peptide bonds—resulting in one or more products. Protein substrates are widely used in research settings as tools in activity assays and high-throughput screening platforms.
