Overview

SEC61B (Protein transport protein Sec61 subunit beta) is a single-pass integral membrane protein that forms part of the heterotrimeric Sec61 complex, the central translocon channel of the endoplasmic reticulum (ER) membrane. The Sec61 complex, comprised of alpha, beta (SEC61B), and gamma subunits, catalyzes the cotranslational and posttranslational translocation of nascent secretory and membrane proteins across or into the ER membrane[1][2]. SEC61B helps maintain the structure and stability of the Sec61 channel but is not directly involved in channel gating or substrate specificity; its deletion attenuates, but does not abolish, Sec61 function in many organisms[2]. Loss-of-function or disruption of the Sec61 complex leads to defects in protein biogenesis, ER stress, and potentially broad cellular dysfunction. There are currently no approved drugs directly targeting SEC61B, nor is it used clinically as a biomarker[2][3].
