Overview

Protein transport protein Sec61 subunit gamma (SEC61G) is an essential single-pass membrane protein that forms part of the Sec61 heterotrimeric channel complex, which mediates the translocation of newly synthesized proteins across and into the endoplasmic reticulum (ER) membrane in eukaryotic cells[1][3][5]. SEC61G functions alongside the alpha (SEC61A1) and beta (SEC61B) subunits, stabilizing the complex by acting as a clamp that holds the channel together[3]. The Sec61 translocon is critical for the co- and post-translational import of secretory and membrane proteins, integrating with ribosomes and various accessory factors, and is implicated in processes such as the unfolded protein response and ER-associated degradation (ERAD)[1][3]. Mutations or altered function in SEC61G or the Sec61 complex are associated with diverse diseases, including certain cancers, by disrupting protein homeostasis and ER signaling[1]. The Sec61 channel, including SEC61G, is also recognized as a potential therapeutic target for modulating protein import into the ER[1][3]. No direct therapeutic drugs against SEC61G are currently in use or clinical development, but research continues on the pharmacological targeting of the Sec61 complex in disease contexts.
