Protein-tyrosine kinase 6 (PTK6), also known as Breast tumor kinase (BRK) or Src-related intestinal kinase (SIK), is a non-receptor, intracellular tyrosine kinase distantly related to the SRC family kinases. It is primarily expressed in epithelial tissues and functions as an intracellular signal transducer, regulating cell differentiation, cell cycle progression, adhesion, survival, and proliferation. PTK6 plays a role in cancer, particularly in breast, prostate, and colorectal cancers, and is being investigated as a potential therapeutic target. Phosphorylation at Y342 activates the enzyme, while phosphorylation at Y447 inhibits activity.
