Protein Tyrosine Phosphatase 1B (PTP1B) is an intracellular non-receptor protein tyrosine phosphatase that acts as a negative regulator in several signaling pathways, most notably insulin receptor signaling. It dephosphorylates phosphotyrosine residues on target proteins, attenuating signal transduction. Inhibitors of PTP1B are being explored as potential therapeutics for type 2 diabetes, obesity, and certain cancers.
PTP1B inhibitors bind to the active site, preventing dephosphorylation of target proteins and thus enhancing insulin signaling and other pathways.
