Overview

SHP2 is a ubiquitously expressed non-receptor protein tyrosine phosphatase that plays a central role in cell signaling. It acts as an intracellular signal transducer and is involved in the regulation of several key cellular processes including cell growth, differentiation, mitotic cycle progression, and oncogenic transformation. SHP2 positively modulates the RAS/MAPK signaling pathway and negatively regulates the PI3K/AKT pathway. Mutations or dysregulation of *PTPN11* are implicated in several diseases, including Noonan syndrome and various cancers. SHP2 has emerged as an important drug target, particularly for solid tumors, and allosteric inhibitors are under clinical investigation.
