Overview

The target is a specific peptide-HLA complex consisting of a fragment of the Protein Tyrosine Phosphatase Receptor Type Z1 (PTPRZ1) protein presented by the Human Leukocyte Antigen (HLA) allele A*02. PTPRZ1 is a transmembrane protein highly overexpressed in glioblastoma, particularly in glioblastoma stem cells (GSCs), where it plays a role in tumor stemness, proliferation, and invasion. The TCT-001 T-cell receptor (TCR) is engineered to specifically recognize this PTPRZ1 peptide–HLA-A*02 complex on the surface of malignant cells. Upon binding, the TCT-001-transduced T cells are activated, releasing cytotoxic granules and cytokines to induce apoptosis in the target tumor cells. This approach aims to provide a highly selective immunotherapy for glioblastoma, a disease with significant unmet medical need and limited effective treatments. PTPRZ1 is considered a clinically relevant target due to its association with tumor stemness and its minimal expression in normal adult tissues outside the central nervous system. The TCT-001 therapy utilizes a non-viral, non-integrating platform for TCR delivery, potentially improving the safety and efficacy of the treatment.

Mechanism of action

TCT-001 is a TCR-engineered T-cell therapy that recognizes the PTPRZ1 peptide presented by HLA-A*02 on the surface of glioblastoma cells, leading to T-cell activation and tumor cell lysis.
