Protein tyrosine phosphatase type IVA 3 (PTP4A3/PRL-3) is a prenylated, dual-specificity, non-receptor protein tyrosine phosphatase that regulates key signaling pathways involved in cell proliferation and migration. It is highly expressed in various cancers, notably metastatic colorectal cancers, T-cell acute lymphoblastic leukemia, and uveal melanoma, where it contributes to tumor progression, invasion, and angiogenesis. Mechanistically, PTP4A3/PRL-3 modulates Src signaling and is involved in the regulation of matrix metalloproteinases (e.g., MMP14), facilitating tumor cell migration and invasion. PTP4A3 is considered a promising therapeutic target, and ongoing research focuses on the development of inhibitory compounds and their application in personalized cancer therapy
