Overview

Protein Wnt-11 (WNT11) is a secreted glycoprotein member of the Wnt family, acting as a ligand for Frizzled transmembrane receptors to mediate non-canonical Wnt signaling pathways (including Wnt/planar cell polarity and Wnt/Ca2+), as well as possible interplay with the canonical beta-catenin pathway[3][6]. WNT11 is essential for embryonic development, regulating tissue patterning, cell fate, cardiac and skeletal morphogenesis, and organogenesis. In adults, WNT11 modulates cell migration, proliferation, and tissue remodeling in both physiological contexts (such as wound healing and regeneration) and pathological conditions, particularly various cancers where its role is highly context- and tissue-dependent: WNT11 may facilitate tumor migration and invasion in some cancers, while inhibiting proliferation and migration in others[1][5][7]. WNT11 expression is induced by signals like TNFα via transcription factors such as EGR1, linking inflammation to dysregulated Wnt signaling in disease[1]. Owing to its central role in development and cancer, WNT11 is an emerging but challenging therapeutic target[5].

Safety considerations

Targeting WNT11 or Wnt signaling may cause developmental toxicity and adverse effects in tissue homeostasis, especially in the heart, skeleton, and other organs in which WNT11 has key developmental or regenerative functions[3][5]. Tumor-promoting and suppressive effects are context-dependent, complicating therapeutic modulation[5].

No approved or common clinical small-molecule or biologic drugs directly targeting WNT11; it is a research target. WNT pathway inhibitors (targeting Frizzled, Porcupine (PORCN), or downstream effectors) may affect WNT11 activity, but WNT11-specific inhibitors are not documented in clinical use[7].
