Overview

Protein Wnt-7a (WNT7A) is a secreted glycoprotein that acts as a ligand within the Wnt signaling cascade, binding Frizzled family receptors to regulate β-catenin-dependent gene transcription and alternative non-canonical pathways. It is essential for embryonic limb patterning, tissue regeneration, neural development, and maintenance of the blood-brain barrier. Mutations disrupt limb and organ development, as seen in syndromes like Fuhrmann. Aberrant WNT7A signaling promotes cancer cell proliferation, and its pathway (often involving FZD7/JAK1/STAT3 or Wnt/β-catenin axes) is a target for investigational therapy. Therapeutic intervention requires balanced modulation due to its role in normal physiology and the potential for wide-ranging side effects
