Protein yippee-like 3 (YPEL3) is a small, evolutionarily conserved protein encoded by the YPEL3 gene, located on chromosome 16p11.2. It belongs to the yippee-like protein family and contains a zinc-finger-like motif. YPEL3 is transcriptionally regulated by p53 and is an important mediator of cellular senescence and apoptosis in both normal and tumor cells. It acts as a tumor suppressor, frequently downregulated in several cancers due to epigenetic silencing. Beyond its growth-inhibitory role, YPEL3 is fundamental for the development of myelinating glial cells—oligodendrocytes and Schwann cells—in the central and peripheral nervous system. Loss or mutation of YPEL3 can lead to defective myelination and peripheral neuropathies. Despite its biological significance, there are currently no approved drugs targeting YPEL3 directly, and its manipulation poses safety concerns due to its essential roles in cell viability and nervous system development[1][2][3][4][5].
