Overview

Protein zwilch homolog (ZWILCH) is an essential component of the ROD-Zwilch-ZW10 (RZZ) complex, which assembles as a fibrous corona structure on the outer kinetochore during mitosis[2][4][9]. This complex is critical for the recruitment of dynein-dynactin motors and spindle assembly checkpoint proteins (MAD1-MAD2) to kinetochores, ensuring correct chromosome-microtubule interactions and regulating the accurate timing of anaphase onset[1][3][4][5][7][9]. ZWILCH is evolutionarily conserved from Drosophila to humans and localizes to kinetochores and kinetochore microtubules during cell division[1][7][8]. Mutations or dysfunction of ZWILCH disrupts kinetochore attachment and spindle checkpoint integrity, thereby promoting lagging chromosomes, aneuploidy, and other mitotic failures—a phenotype linked to cancer development[1][7][9]. The RZZ complex provides an assembly and signaling platform for proteins governing chromosome segregation and spindle assembly checkpoint fidelity.
