Overview

Proteinase 3 (PR3), also known as PRTN3, is a serine protease enzyme primarily expressed in neutrophil granulocytes. It plays a key role in innate immunity, degrades extracellular matrix proteins, and activates inflammatory cytokines. Clinically, it is known as the primary autoantigen targeted by anti-neutrophil cytoplasmic antibodies (ANCAs) of the c-ANCA subtype, highly associated with granulomatosis with polyangiitis (GPA) and other vasculitides. It acts both intracellularly—regulating proliferation or apoptosis—and at cell membranes where its expression correlates with chronic inflammatory risk.
