Overview

The phrase “proteins of necrotic tissue” refers to the heterogeneous group of intracellular and membrane-associated proteins released into the extracellular environment following loss of cell membrane integrity during necrosis, a form of regulated or accidental cell death characterized by cellular swelling and plasma membrane rupture. These proteins do not constitute a single molecular entity or druggable receptor, but are widely used as generic biomarkers of tissue damage or necrosis in pathological settings such as infections, cancer, and ischemic injuries. Unlike proteins from programmed death (apoptosis or necroptosis[1][3]), necrotic tissue proteins generally include a non-specific mix of nuclear, cytosolic, and organelle-derived molecules, many of which can function as DAMPs that promote inflammation and immune activation. Their measurement may help diagnose or monitor diseases with necrotic pathology, but the term is not precise enough to represent a specific target for drug development. No precise, standardized receptor or protein is known as “Proteins of necrotic tissue,” so it does not fit criteria for a molecular drug target.
