Proto-oncogene protein c-bcl2 (Bcl-2) is a key anti-apoptotic protein that promotes cell survival by inhibiting programmed cell death. It primarily localizes to the outer mitochondrial membrane, endoplasmic reticulum, and nuclear envelope. By blocking the activation of pro-apoptotic proteins like Bax and Bak, Bcl-2 prevents mitochondrial outer membrane permeabilization and subsequent caspase activation. Overexpression or dysregulation of Bcl-2 is frequently observed in various cancers, contributing to tumorigenesis and resistance to chemotherapy. As a result, Bcl-2 has become an important therapeutic target with small molecule inhibitors being developed to restore apoptotic sensitivity in cancer cells.
