MDMX (also known as MDM4) is a key negative regulator of the tumor suppressor p53. It inhibits p53's transcriptional activity by binding to its transactivation domain. MDMX cooperates with MDM2 to promote p53 degradation and has p53-independent oncogenic functions. Overexpression of MDMX is observed in various cancers, making it a therapeutic target for reactivating p53.
