Proto-oncogene protein Wnt-3 (Wnt3) is a highly conserved, secreted signaling ligand belonging to the Wnt family of morphogens. It plays a central role in the activation of the canonical Wnt/β-catenin pathway, mediating cell fate determination, proliferation, and differentiation during embryonic development. Wnt3 binds to Frizzled and LRP5/6 membrane receptors, triggering a cascade that stabilizes β-catenin and drives transcription of Wnt target genes. Dysregulation or overexpression of Wnt3 is implicated in the initiation, progression, and resistance to therapy in several human cancers. Targeting Wnt3-mediated signaling is an area of therapeutic interest, though challenges remain due to its fundamental roles in normal tissue homeostasis and development.
