Overview

Protocadherin beta-11 is encoded by the *PCDHB11* gene, part of a larger beta-cluster of protocadherins located on chromosome 5. Unlike the classical cadherins, beta-protocadherins have a single large exon encoding six extracellular cadherin domains, a transmembrane domain, and a unique cytoplasmic tail. These proteins form homophilic interactions on neuronal cell surfaces, believed to mediate neural circuit specificity by regulating cell-cell adhesion. Human PCDHB11 exhibits unusually high numbers of human-specific amino acid changes with low intra-human variability, suggestive of rapid evolutionary divergence but no major change in function found to date[1][3][4][5]. Its primary known or proposed function is to facilitate the correct wiring of neuronal networks in the central nervous system, potentially influencing cognitive function and disease susceptibility[1][3]. There are reported associations with autism spectrum disorder, though no confirmed mechanistic connections or therapeutic relevance. If additional, structured or clinical info is required, PCDHB11 would currently yield mostly unvalidated hypotheses owing to limited functional characterization and absence of direct drug interactions.
