Overview

Protocadherin beta-2 is a neural cadherin-like cell adhesion protein encoded by the PCDHB2 gene in humans, belonging to the protocadherin beta gene cluster on chromosome 5. These proteins are integral plasma membrane proteins that predominantly occur in the brain and are thought to play a critical role in the establishment and function of specific cell-cell neural connections. Protocadherin beta-2 is part of the cadherin superfamily, containing six extracellular cadherin domains and a cytoplasmic tail with a distinct structure compared to classical cadherins. The extracellular domains mediate homophilic (self-specific) cell-cell adhesion, likely contributing to the diversity and specificity of neuronal wiring during development. Mutations or disruptions in PCDHB2 have been associated with neurodevelopmental disorders such as autism spectrum disorder, though its specific functions and potential as a therapeutic target are not clearly established at present. There is currently no evidence that Protocadherin beta-2 is directly targeted by therapeutic drugs, nor is it used as a biomarker or known to present clinical safety concerns related to therapy. While defects in related protocadherins have been linked to cancer and neuropsychiatric disease, the primary role of Protocadherin beta-2 is in neural cell adhesion and circuit assembly, not as a classical therapeutic receptor, enzyme, or transporter.
