Overview

Protocadherin beta-9 is a neural cadherin-like cell adhesion protein encoded by the PCDHB9 gene, part of the protocadherin beta cluster on chromosome 5[1][2][3]. The protocadherin beta family is composed of integral plasma membrane proteins with six extracellular cadherin domains and a unique cytoplasmic tail[1][2]. These proteins mediate homophilic (like-with-like) cell-cell adhesion, likely specifying differential neural connections critical for establishment and function of precise neuronal circuits[1][2][3]. The protocadherin beta cluster's organization resembles immune receptor gene clusters, and each beta isoform is encoded by its own large exon rather than sharing 3' exons like alpha and gamma clusters[1][2]. Protocadherin beta-9 is expressed mainly in the brain and central nervous system and probably contributes to neuronal connectivity and survival during development[1][2][3][4]. Although the exact functions of Protocadherin beta-9 remain to be fully defined, disruption of protocadherin gene clusters has been linked to neurodevelopmental disorders, including autism spectrum disorder[1]. No drugs or biomarkers are currently known to target protocadherin beta-9 directly.
