Overview

Protocadherin gamma-A6 (PCDHGA6) is a calcium-dependent cell-adhesion protein, a member of the protocadherin gamma gene cluster within the larger cadherin superfamily[1]. It is primarily expressed in the brain, where it is believed to play critical roles in the establishment and function of specific cell-cell connections among neurons[1][2][3]. The gamma cluster, to which it belongs, is characterized by an immunoglobulin-like genomic organization, and produces neural cell adhesion proteins with six extracellular cadherin domains, a transmembrane region, and a common cytoplasmic region[1][2][3]. These molecules engage in both homophilic trans interactions (between neighboring cells) and promiscuous cis interactions (on the same cell), providing remarkable surface diversity critical for neuronal self-recognition and wiring specificity[2][3][4]. PCDHGA6 has been associated with neurodevelopmental disorders such as microcephaly and disorders marked by poor growth and skeletal anomalies[1]. There are currently no approved drugs, established mechanisms of action, or well-defined safety concerns directly related to therapeutically targeting PCDHGA6[1].
