Overview

Protocadherin gamma-B5 (PCDHGB5) is a neural cadherin-like, calcium-dependent cell adhesion protein that is part of the protocadherin gamma gene cluster on chromosome 5[1][2][6]. This gene cluster is thought to play a pivotal role in forming and maintaining specific cell-cell connections in the brain, particularly involved in neuronal connectivity. The protein contains six extracellular cadherin domains, a transmembrane region, and a shared cytoplasmic domain (with other gamma cluster members) involved in intracellular signaling. Although it has important roles in brain development and function, there is currently no evidence that it serves as a direct therapeutic target, is associated with drug interactions, or is used as a biomarker or for patient selection[1][5][6]. Diseases linked to mutations in PCDHGB5 include certain neurodevelopmental disorders and digenic forms of facioscapulohumeral muscular dystrophy[1].
