Overview

Protocadherin gamma-B7 (PCDHGB7) is a neural cadherin-like cell adhesion protein encoded by the PCDHGB7 gene, located within the protocadherin gamma gene cluster on chromosome 5[1][2]. It is part of the gamma subfamily B cluster and likely plays a critical role in the establishment and maintenance of neuron-specific cell–cell connections in the brain, contributing to neural circuit assembly and synaptic specificity[1][2]. It has multiple extracellular cadherin domains and a transmembrane domain, with a constant cytoplasmic region, and is mainly expressed in neural tissues[5]. In cancer, PCDHGB7 is frequently hypermethylated and downregulated, functioning as a tumor suppressor by inducing cell cycle arrest and apoptosis. The hypermethylation status of PCDHGB7 is a robust biomarker for the early, minimally invasive detection of several cancers, particularly endometrial cancer, outperforming conventional markers such as serum CA125[3].
