Protocadherin gamma-C5 is a member of the protocadherin gamma gene cluster, belonging to the cadherin superfamily of calcium-dependent cell-adhesion proteins[1][6]. It is encoded by the PCDHGC5 gene, located on chromosome 5[1][4]. The protein consists of six extracellular cadherin domains, a transmembrane region, and a cytoplasmic tail common to the gamma cluster[1][2]. Protocadherin gamma-C5 is primarily expressed in the brain, where it is thought to play a critical role in establishing and maintaining specific neuronal connections via homophilic and heterophilic cell–cell adhesion mechanisms, contributing to neuronal self-recognition and the diversification of neuronal circuitry[1][2][6]. Mutations in PCDHGC5 have been associated with intellectual developmental disorder and developmental and epileptic encephalopathy[1]. There are currently no known drug interactions or direct therapeutic targeting of this molecule[1][6].
