Overview

Proton-coupled amino acid transporter 2 (SLC36A2) is a membrane protein that functions as a pH-dependent proton-coupled symporter, selectively transporting small amino acids such as glycine, alanine, and proline across cellular membranes. It is electrogenic and exhibits high selectivity for amino acids with small side chains, preferring both L- and D-enantiomers. Expression is notable at the apical membrane of the renal proximal tubule and in the plasma membrane of brown/beige adipocytes, where it also acts as an extracellular amino acid sensor to regulate lysosomal acidification. Mutations in SLC36A2 are associated with inherited metabolic disorders such as iminoglycinuria and hyperglycinuria[1][2][5]. No evidence currently supports its direct targeting by approved or investigational drugs, or its established use as a biomarker or major safety concerns in therapeutic contexts.
