The proton-coupled folate transporter (PCFT), encoded by the SLC46A1 gene, is a membrane protein responsible for the absorption of folates in the small intestine and delivery to the central nervous system. It functions as an electrogenic proton symporter, utilizing a proton gradient to transport folates into cells. PCFT is a target for developing antifolate chemotherapeutics that selectively enter cancer cells, which often have acidic microenvironments, to reduce toxicity.
