Overview

The Pseudomonas aeruginosa lipopolysaccharide O-polysaccharide of serotype O11, also known as the O11 O-antigen, is a critical component of the bacterial outer membrane and a major virulence factor [8, 15]. It consists of repeating trisaccharide units—specifically [-3)–α-L-FucNAc–(1-3)–β-D-FucNAc–(1-2)–β-D-Glc-(1-)]—that extend from the cell surface, providing a physical barrier and conferring resistance to host serum-mediated killing [8, 17]. Serotype O11 is one of the most prevalent serotypes identified in clinical settings, particularly in hospital-acquired and ventilator-associated pneumonia [1, 11]. As an immunodominant surface antigen, it serves as a primary target for the development of targeted immunotherapies and vaccines [4, 16]. Drugs such as Panobacumab (AR-101), a fully human IgM monoclonal antibody, specifically bind to this O-polysaccharide to promote opsonophagocytic killing and complement-dependent bactericidal activity [2, 5]. While highly effective against the O11 serotype, the narrow specificity of such treatments necessitates rapid diagnostic identification of the infecting strain's serotype [10, 12].
