Overview

The Pseudonaja textilis prothrombin activator complex (Pt-PA) is a potent enzymatic toxin found in the venom of the Eastern Brown Snake (Pseudonaja textilis). It is a multi-protein complex structurally and functionally analogous to the human prothrombinase complex, consisting of a Factor Xa-like serine protease and a Factor Va-like cofactor (UniProt: P81428, P60591). Unlike the human counterpart, Pt-PA is constitutively active and does not require phospholipids or calcium for its primary function, allowing it to rapidly convert host prothrombin into thrombin (PubMed: 12606768). This uncontrolled thrombin generation leads to the rapid exhaustion of fibrinogen and other clotting factors, a condition known as venom-induced consumption coagulopathy (VICC) (PubMed: 16131498). In a clinical setting, Pt-PA is the primary target for neutralization by specific antivenoms to prevent life-threatening hemorrhage in snakebite victims (PubMed: 24635461). The complex's ability to bypass normal regulatory checkpoints of the coagulation cascade makes it one of the most lethal components of brown snake venom (PubMed: 15130611).

Specific antivenom antibodies bind to the subunits of the prothrombin activator complex, sterically hindering its enzymatic site and neutralizing its ability to convert prothrombin into thrombin (PubMed: 24635461).
