Psl exopolysaccharide (Psl)

Target
Psl
Molecular classification
Exopolysaccharide, Biofilm matrix component, Polysaccharide matrix factor
01

Overview

Psl exopolysaccharide is a neutral exopolysaccharide synthesized by the *pslA-O* operon in *Pseudomonas aeruginosa*. It is essential for initial surface attachment and the maintenance of biofilm microstructure, providing mechanical stability and protection against antimicrobials and the host immune system. Psl is composed primarily of mannose, galactose, glucose, and rhamnose in a pentasaccharide repeating unit and is a major target for strategies aiming to prevent or disrupt biofilm-mediated infections, especially in cystic fibrosis and hospital-acquired infections[1][3][4][6][7].

02

Mechanism of action

Agents targeting Psl may inhibit exopolysaccharide synthesis (e.g., inhibitors of the Psl synthetic operon), disrupt established biofilm matrices, or enhance susceptibility of bacteria to antibiotics.

05

Safety considerations

Therapeutic targeting of Psl exopolysaccharide primarily affects microbial biofilms and is not expected to have direct toxicity in humans, but disruption of biofilms may result in the release of bacterial cells and toxins, potentially worsening acute infection.
06

Biomarkers

Psl presence or quantification may serve as a marker for biofilm-forming strains of *P. aeruginosa*
