PSMB8 antisense RNA 1 (PSMB8-AS1) is a long non-coding RNA (lncRNA) located near the PSMB8 gene, transcribed in an antisense orientation[2][1]. It is predominantly nuclear and regulates gene transcription through interactions with transcription factors and chromatin[1][2]. In cardiovascular biology, PSMB8-AS1 is significantly upregulated in atherosclerotic plaques and in the serum of patients with coronary heart disease, where it promotes vascular inflammation, monocyte/macrophage adherence to endothelium, and atherosclerosis development by recruiting the transcription factor NONO to the promoter of PSMB9, which upregulates inflammatory adhesion molecules like VCAM1 and ICAM1[1]. PSMB8-AS1 is also induced by influenza virus infection and type I interferon in various epithelial and kidney cells; knockdown of PSMB8-AS1 reduces viral replication, indicating a pro-viral function in infection[2]. The lncRNA does not currently have any approved drugs directly targeting it, nor is it a conventional therapeutic target (receptor, enzyme, transporter). Its main significance is as a disease biomarker and as a basic research target for understanding gene regulation in inflammation and infection.
